Effect of Vernonia amygdalina on phospholipase activity from Naja Mossambica (Cobra)

Background: Phospholipases are one of the numerous enzymes found in Naja mossambica venom. They play a major role snakebite envenomation, and also responsible for hydrolysis phospholipid, disrupting membrane integrity. In this study, we evaluated effect Vernonia amygdalina on Phospholipase activity from (Cobra) Results: Partially purified phospholipase had maximal velocity (Vmax) Michaelis Menten constant (Km) 7.6 × 10-5 mol/min 1.7mg/ml, while crude Vmax Km 9.4 10-5mol/min 2.5mg/ml respectively. Inhibition study aqueous extracts leaf shows that extract is potent inhibitor dose-dependent pattern. The different doses 15 %, 10 % 5% produced percentage inhibition 74.04 78.6 86.63% kinetic binding (Ki) values concentrations 5%, 10% 15% were 0.21mg/ml, 0.29mg/ml 0.39mg/ml, partially 0.48mg/ml, 0.42mg/ml 0.41mg/ml It can be deduced results inhibits an uncompetitive manner. Conclusion: Aqueous leaves may contain some bioactive agents could serve as inhibitors
